Today we began to culture our yeast on each medium. On solid mediums we add 15uL of yeast were added to each plate. 15uL of WT yeast was added to three plates of YPD and to three plates of YPE agar. 15 uL of KO yeast was added to three plates of YPD agar and three plates of YPE agar.

      Once the yeast was added to the solid agar plates we began to dilute both yeast strains (WT and KO) to obtain a total concentration of 1/32.

     When the yeast was diluted to our desired concentration we added  it to our liquid culture. We had three 1.5mL tubes filled with 1mL of YPD liquid medium and three 1.5mL tubes filled with 1mL of YPE liquid medium. We added 250uL of the diluted WT and KO to each 1.5mL tubes.

We incubated each culture for five days.
