In today's class we will:

-redo the paper based assay because contamination grew

-use a different technique

  -used a ziplock bag as a humidity chamber

  -added ethanol and switch it around to the bag and dry the bag.

  -Sterilize the paper with ethanol, let it dry. then, soak the paper in sterile water, then place the paper in the bag.

  -Place Petri-dish with shapes tag yeast on top of the paper in the bag and incubate it. 

The optimal density of yeast: mutant=0.93

           wild type=0.97
