Transformation of Yeast- 09/12/16

Protocol

This is the protocol we followed to transform the yeast. The class started at the second half of step 3. However, in step 10, instead of resuspending the yeast in ddH2O we used a nutrient rich medium, YPD (200μL).

Before class we were required to read this paper by Gietz and Woods regarding yeast transformation

Calculations

To calculate the amount of water we would need to dilute the LiAc we used:

(1MLiAc)(10μLddH2O)=(0.1M)(V)(2)

Observations

The only observation our group made was that the pellet was a white solid-like substance.
